چکیده

2006 I hereby declare, that thesis has been written independently and with no other sources and aids than quoted. the lab and for such excellent technical assistance throughout my PhD. Many thanks also to Kittel and Wernher Fouquet for helping whenever needed, for lending me your ears in case of problems and for answering " stupid " questions. Many thanks also to Miriam Richter, Jasmin Held and Heiko Röhse for technical assistance. Additionally, I want to thank all ENI-members, including the ones of " ENI-classic " , for creating the unique " young " atmosphere at the ENI. I am specially grateful to my " scientific advisor " Mika Ruonala for his criticism, help, advise and friendship throughout all my PhD. Many thanks go also to Aspi and Carlos Merino for their optimism and fruitful talks about " science and life " exactly when I needed to be cheered up. I also want to thank Frank, Dirk, and Magda for providing practical help whenever needed.

Aconitase and Developmental EndPointsasEarly IndicatorsofCellularToxicity Induced by Xenobiotics in Drosophila Melanogaster

Background: In this study, the toxicity of the different xenobiotics was tested on the fruit fly Drosophila melanogaster model system.  Methods: Fly larvae were raised on food supplemented with xenobioticsat different concentrations (sodium nitroprusside (0.1-1.5 mM), S-nitrosoglutathione (0.5-4 mM), and potassium ferrocyanide (1 mM)). Emergence of flies, food intake by larvae, and pupation h...

Concentration dependent effect of morphine, aspirin, capsaicin and chili pepper hydro alcoholic extract on thermal and chemical pain model in fruit fly (Drosophila melanogaster)

Introduction: Pain research using animal models is related to ethical concerns, so invertebrates and insects have been recommended by researchers. In the present study, the nociceptive and antinociceptive effects of capsaicin, aspirin, morphine and chili extract were examined using fruit fly (Drosophila melanogaster) as an alternative for rodent pain model. Methods: Stage 3 of larvae and ad...

Toxicological Evaluation of a New Lepidopteran Insecticide, Flubendiamide, in Non-Target Drosophila melanogaster Meigen (Diptera: Drosophilidae)

Background: Flubendiamide, comparatively a new pesticide designed to eradicate lepidopteran insect pests is known to have low risk to birds, mammals, fish, algae, honey bees, non-target arthropods, earthworms, soil macro- and micro-organisms, non-target plants as well as sewage treatment organisms; however, the risk assessment for aquatic invertebrates from metabolite could not be finalized wit...

Arg206 of SNAP-25 is essential for neuroexocytosis at the Drosophila melanogaster neuromuscular junction.

An analysis of SNAP-25 isoform sequences indicates that there is a highly conserved arginine residue (198 in vertebrates, 206 in the genus Drosophila) within the C-terminal region, which is cleaved by botulinum neurotoxin A, with consequent blockade of neuroexocytosis. The possibility that it may play an important role in the function of the neuroexocytosis machinery was tested at neuromuscular...

Conservation of the protein composition and electron microscopy structure of Drosophila melanogaster and human spliceosomal complexes.

Comprehensive proteomics analyses of spliceosomal complexes are currently limited to those in humans, and thus, it is unclear to what extent the spliceosome's highly complex composition and compositional dynamics are conserved among metazoans. Here we affinity purified Drosophila melanogaster spliceosomal B and C complexes formed in Kc cell nuclear extract. Mass spectrometry revealed that their...

Human stoned B interacts with AP-2 and synaptotagmin and facilitates clathrin-coated vesicle uncoating.

Synaptic vesicle biogenesis involves the recycling of synaptic vesicle components by clathrin-mediated endocytosis from the presynaptic membrane. stoned B, a protein encoded by the stoned locus in Drosophila melanogaster has been shown to regulate vesicle recycling by interacting with synaptotagmin. We report here the identification and characterization of a human homolog of stoned B (hStnB). H...
